1. Title: it need to re-write to be more clear, concise and informative.

2- The authors would add more information about the methodology in the abstract.

3. The graphical abstract is highly recommended.

4. The authors would add the originality of their work.

5- The authors would add some results of docking analysis in the abstract.

6- Please add more information about Cyperus conglomeratus in the introduction.

7- “According to research studies, the underlying mechanisms of prebiotics effects on these disorders are through regulating the composition of the microbiota and its metabolites, in addition to improving the functions on the intestinal mucosal barrier” Rewrite please.

8- The authors would add more references in the methodology part.

9- Kindly, could you recheck the English grammar, the syntax and spaces between words?

The manuscript entitled " Inhibition of α-Amylase, α-Glucosidase, and Glycogen Phosphorylase by Aurone Derivatives and Their Prebiotic Activities" contains very important data, as they isolated three molecules from the Cyperus conglomeratus plant and determined their biological activities. I have some suggestions for authors to make their manuscript clearer for readers.

1.      Rephrase ” Cyperus conglomeratus has been utilized in traditional medicine as an emollient, diuretic, analgesic, anthelmintic, and for the treatment of some diseases.” to be “Cyperus conglomeratus has been utilized in traditional medicine as an emollient, diuretic,
analgesic, anthelmintic, and other diseases.”

2.      To the abstract part add to used methods and apparatuses you used in the identifications of compounds and bio-tests.

3.      At the end of the abstract add kindly your conclusion and future recommendations.

4.      Add suitable citations to the first sentence of the (1. Introduction).

5.       Add suitable citations to the “Thus, glycogen phosphorylase inhibitors attracted specific attention because they might prevent glycogenolysis at high blood glucose levels in type II diabetes.

6.      At the end of the introduction part add the aims of your study.

7.      Add to the 2.1. Plant material the plant voucher specimen code and where the herbarium was deposited also the methods of drying.

8.      At the end of the material and methods part, add the statistical analysis section.

9.      The results and discussion parts are well described.

10.   At the end of the conclusion part add your own recommendations for future work on these compounds

In this manuscript of "Inhibition of α-Amylase, α-Glucosidase, and Glycogen Phosphorylase by Aurone Derivatives and Their Prebiotic Activities", this study demonstrates the importance of Cyperus conglomeratus as a source of aurone derivatives with diverse biological activities. The findings have implications for the development of new therapeutic agents for diabetes and highlight the potential prebiotic benefits of these compounds on gut health. A lot of work was done and it is contributing to the current scientific knowledge base in some way. But this MS needs revision and the author reply comments properly. The comments and questions are as follows:

 

1. In general, there is a lack of explanation of why authors did these various experiments in the introduction. 

 

2. Relative information about aurone derivatives should be provided in the introduction.

 

3. Has the usage of aurone derivatives as a prebiotic been reported in any other publications before? How do the findings of this study compare to those of earlier research?  

 

4. Please check the format of Table 1. .

 

5. Figure 2. is showing as an incomplete image; kindly consider replacing it with a complete image.
