The ETS Transcription Factor Family in Human Diseases

Special Issue Information

Dear Colleagues,

Forty years after the identification of the original vETS gene in the E26 virus and the entire ETS gene family, it is now believed that they are involved in multiple developmental processes within the RTK pathway and can trigger various forms of cancer, especially prostate cancer, Ewing's sarcoma and lymphomas. In spite of the extensive amount of published work, there are still many unclear points that could allow for a better understanding and utilization of the family members. For example, the mechanism of transcriptional activation or repression and any possible differences when acting proximal or distal to the start site, as well as cross-talk with other pathways. An important underlying question given the co-expression of multiple family members in most of the cells and the sharing of a wide range of DNA binding sites  is the cooperation and co-dependence among different family members and the extent that the various identified phenotypes are gene autonomous or the result of an ETS gene imbalance. Finally, although the pharmacological targeting of transcription factors remains a challenging task, further efforts in this direction are needed, especially for genes with an established causal effect in diseases or development.
