Dear Colleagues,
Protein aggregates and dysfunctional organelles are the most common features that alter cell homeostasis and trigger neurodegeneration. Compelling evidence indicates that complex 1 of the mammalian target of rapamycin (mTORC1) is a central cell-growth-regulating kinase implicated in multiple essential processes, including cell growth, protein synthesis, and the development as well as function of the nervous system. The induction of mTORC1 activates ribosomal biogenesis and negatively regulates autophagy, which is a lysosomal degradation process of eukaryotic cells to maintain cytoplasmic quality control by clearing cytotoxic proteins and damaged organelles. Moreover, mTORC1 signaling is linked to oxidative stress and brain disorders. Herein, stimulating autophagy contributes to neuroprotection by reducing oxidative stress, representing a new therapeutic approach in the prevention and treatment of neurodegenerative diseases. The above being the case, there is increasing interest in the study of the biological properties of natural products that could act as neuroprotective agents in several neurodegenerative diseases.
