Dear Colleagues,
Proteins are fascinating molecular machines with an extraordinary ability to perform a myriad of functions in vivo. Indeed, proteins are dynamic molecules that can undergo a wide range of motions that are biologically relevant at different levels. Thus, rapid atomic fluctuations, which occur within the femtosecond to picosecond timescale, are essential for processes such as enzyme catalysis. On a larger scale, they can also experience conformational changes and domain rearrangements, from nanoseconds to milliseconds, which are frequently triggered by binding to specific interactors in signal transduction or molecular recognition processes. In this context, the study of the dynamic properties of proteins that contribute to allosteric activation, altering both regulation and substrate specificity, is highly relevant. Particularly noteworthy is allosteric signal transmission in intrinsically disordered proteins, which play a crucial role in cellular regulation and are implicated in various diseases. In general, the association between protein dynamics and conformational diseases has emerged as a critical area of biomedical research. The dysregulation of protein dynamics can disrupt cellular processes, contributing to the onset of various diseases such as neurodegenerative disorders, cancer, and metabolic syndromes; this currently comprises an intense area of research.
This Special Issue of IJMS aims to elucidate protein dynamics and their role in in vivo function/dysfunction. The scope of this Special Issue includes enzyme catalysis, conformational changes, allosteric regulation, signal transduction, and the impact of protein dynamics on diseases such as neurodegenerative disorders and cancer. Additionally, computational methods for studying protein dynamics and potential therapeutic interventions are explored. As Guest Editors, we would like to invite you contribute original papers and reviews to this Special Issue, entitled “Protein Dynamics, Binding and Allostery”.
