Assays for the Impact of Functional Biomaterials on Cell Proliferation, Cell Viability, and Cell Metabolic Activities

Special Issue Information

Dear Colleagues,

Functional biomaterials have been synthesized and advocated for diverse applications as antimicrobial/antiviral agents, medical textiles, hygienic products, medical device coatings, scaffolds for tissue engineering, etc. As the first prerequisite for any intended application, “in vitro” toxicity testing by reliable methods must be conducted to assess the potentially harmful effects of functionalized biomaterials. This Special Issue invites review and research papers that cover traditional and emerging assays of cell proliferation, cell viability, and cell-cycle analysis after cells are exposed to functional biomaterials. As functional biomaterials bind to the cellular membrane or other components, improved methods to overcome possible interferences of functionalized biomaterials with common toxicity assays are also considered. The Special Issue will cover the following topics:

  • Novel fluorescent dyes beyond propidium iodide and 7-aminoactinomycin D that bind to double-stranded DNA or RNA of cells with compromised plasma membranes.
  • Enzymatic activity fluorogenic substrates: substrates that react with intracellular enzymes in live cells, e.g., esterases or extracellular glucose-6-phosphate dehydrogenase and lactate dehydrogenase released from damaged cells.
  • Chemiluminescent ATP assays and other biomarkers.
  • Combined substrates and techniques for probing plasma and intracellular cell membrane integrity (dead cells, stressed organelles such as endoplasmic reticulum (ER) and mitochondria), and live cells (esterases).
  • Metabolic activity agents: Novel agents that can determine the cellular redox potential in live cells. A classic example is the reduction of resazurin by live cells to a red and highly fluorescent resorufin.  
  • Assays using membrane integrity dyes for early apoptosis, e.g., the detection of phosphatidylserine expression on the cell membrane. In this context, this Special Issue also covers the topic of probing biomaterial–cell interactions by spectroscopy (FTIR, Raman, etc.) and spectroscopy (SEM, AFM, etc.)
  • Mitotoxicity agents can be used to detect changes in mitochondrial membrane potential.

The Special Issue also calls for papers related to emerging non-invasive and continuous methods for cell viability assays, e.g., cell-based impedance spectroscopy.
