Dear Colleagues,

The use of antimicrobials as growth promoters and the incorrect use of antibiotics have led to the onset of the worrying phenomenon of antibiotic resistance. Recent estimates report that bacteria resistant to antibiotics are responsible for approximately 33,000 death-causing infections (Cassini et al. 2019) in the EU.
To counteract this dangerous problem, institutions have imposed a reduction in the use of antimicrobials and dietary supplementation with high dosages of zinc oxide (EMA, 2016; EFSA, 2016).
Together with correct management, prevention and biosecurity plans, alternative feed strategies are needed to support pig’s growth and health throughout the breeding phase.
Therefore, this Special Issue welcomes submissions, including reviews or original research studies, on pig nutrition regarding all feeding strategies useful to minimize the use of antimicrobials and zinc oxide.

In particular, the papers may concern, but are not limited to, the use of the following in pig nutrition:

  • acidifiers;
  • probiotics;
  • prebiotics;
  • substances with immunomodulating activity;
  • enzymes;
  • botanicals, vegetal extracts and essential oils;
  • functional feeds;
  • and any nutritional strategy that allows to reduce/eliminate the use of antimicrobials and zinc oxide.

The study was conducted to investigate the effects of replacing antibiotic growth promoters (AGPs) with an egg immunoglobulin (IgY) combined with phytomolecules (PM) on the growth rate, serum immunity, and intestinal health of weaned pigs challenged with Escherichia coli K88 (E. coli K88). A total of 192 piglets were weaned at 28 days old with an average weight of 7.29 (± 0.04) kg. They were randomly divided into four treatments containing eight replicates with six piglets per replicate. The treatment groups were NC and PC fed a basal diet, AGP fed a basal diet supplemented with 75 mg/kg chlortetracycline, 50 mg/kg oxytetracycline calcium, and 40 mg/kg zinc bacitracin, IPM fed a basal diet supplemented with IgY at dose of 2.5 g/kg and 1.0 g/kg and PM at dose of 300 mg/kg and 150 mg/kg during days 1 to 17 and 18 to 42, respectively. On days 7 to 9 of the experiment, piglets in the PC, AGP, and IPM groups were orally challenged with 20 mL E. coli K88 (109 CFU/mL), while piglets in the NC group were challenged with 20 mL medium without E. coli K88. The E. coli K88 challenge model was successful as the incidence of post-weaning diarrhea (PWD) of piglets challenged with E. coli K88 was significantly higher than that of those unchallenged piglets during the challenge time (days 7 to 9) and days 1 to 7 of post-challenge (p < 0.05). A diet with combinations of IgY and PM and AGPs significantly decreased the incidence of PWD during the challenge time and days 1 to 7 of post-challenge (p < 0.05) compared to the PC group and significantly improved the ratio of feed to weight gain (F:G) during days 1 to 17 of the experiment compared to the NC and PC groups (p < 0.05). In comparison with the PC group, piglets in the IPM group had significantly higher serum levels of IgA, IgG, and IgM (p < 0.05), but lower serum IL-1β on day 17 of experiement (p < 0.05). Besides, diet supplementation with AGP significantly decreased serum IL-1β, IL-6, and TNF-α on days 17 and 42 (p < 0.05) with comparison to the PC group. Piglets in the IPM group showed a significantly lower level of fecal coliforms (p < 0.05), but a higher villus height of jejunum and ileum and higher ratio of villus height to crypt depth of duodenum and jejunum (p < 0.05) than those piglets in the PC group. In summary, diet supplementation with a mixture of IgY and PM decreased the incidence of PWD and coliforms, increased feed conversion ratio, and improved intestinal histology and immune function. Early-life microbial colonization is an important driver for the development and maturation of the gut. The present study aimed to determine whether a single-dose supplement given only at birth would improve piglet performance and modify their fecal microbiota during the suckling and post-weaning periods. At birth, piglets from eight litters received a supplement (SUP+) while piglets from six other litters received water (SUP−). All piglets were monitored until two weeks post-weaning, and fecal samples were collected on Day 16 of age and two weeks post-weaning (Day 39 ± 1). The supplementation resulted in an improvement of average daily gain during the whole experimental period, mainly due to a better growth and a reduction in the incidence of diarrhea in the post-weaning period. There were no differences in the abundance and diversity of the main taxa, although the supplementation increased the relative abundance of rare taxa, such as bacteria from the Saccharibacteria and Cyanobacteria phyla, and the Lentisphaeria class in the suckling period. In addition, at 16 days of age, SUP+ piglets had a more diverse core microbiota, with bacteria from the Lactobacillus genus being present in the core microbiota of SUP+ piglets and absent from SUP− piglets. Therefore, the enhanced growth performance and reduction in diarrhea seem to be related to changes in fecal microbiota during the suckling period rather than at two weeks post-weaning. Full article

This study investigated the effects of Clostridium butyricum (C. butyricum) use on growth performance, serum immunity, intestinal morphology, and microbiota as an antibiotic alternative in weaned piglets. Over the course of 28 days, 120 piglets were allocated to four treatments with six replicates of five piglets each. The treatments were: CON (basal diet); AGP (basal diet supplemented with 0.075 g/kg chlortetracycline, 0.055 g/kg kitasamycin, and 0.01 g/kg virginiamycin); CBN (basal diet supplemented with normal dosage of 2.5 × 108 CFU/kg C. butyricum); and CBH (basal diet supplemented with high dosage of 2.5 × 109 CFU/kg C. butyricum). Body weight (BW) and feed consumption were recorded at the beginning and on days 14 and 28 of the experiment, and representative feed samples and fresh feces were collected from each pen between days 26 and 28. Average fecal score of diarrhea was visually assessed each morning during the experimental period. On the morning of days 14 and 28, blood samples were collected to prepare serum for immune and antioxidant parameters measurement. One male piglet close to the average group BW was selected from each replicate and was slaughtered on day 21 of the experiment. Intestinal crypt villi, and colonic microbiota and its metabolites short-chain fatty acids were measured. Compared to the CON group, the CBN and AGP groups significantly decreased (p < 0.05) the ratio of feed to weight gain by 8.86% and 8.37% between days 1 and 14, 3.96% and 13.36% between days 15 and 28, 5.47% and 11.44% between days 1 and 28. Dietary treatment with C. butyricum and AGPs significantly decreased the average fecal score during the experimental period (p < 0.05). The apparent total tract digestibility of dry matter, organic matter, and total carbohydrates in the CBH group were higher respectively at 3.27%, 2.90%, and 2.97%, than those in the CON or AGP groups (p < 0.05). Compared to the CON group, the CBH group significantly increased short-chain fatty acids in colon and villus height in the jejunum (p < 0.05). The CBN group had higher serum levels of immunoglobulins, interleukin 2 (IL-2), and glutathione peroxidase (GSH-PX) activity, but lower serum levels of IL-1β and IL-6, and a lower aspartate aminotransferase (AST), alkaline phosphatase (ALP), and gamma-glutamyl transpeptidase (γ-GT) activity (p < 0.05), while compared to the CON group. Dietary treatment with C. butyricum significantly increased the relative abundance of Streptococcus and Bifidobacterium (p < 0.05). In summary, diet with C. butyricum increased the growth performance and benefited the health of weaned piglets. Full article

Benzoic acid (BA) and essential oils (EOs) (thymol, 2-methoxyphenol, eugenol, piperine, and curcumin) are being studied to minimize the impairment of gastrointestinal functions in weanling piglets. This study evaluates the effects of combining BA and EO on the performance and intestinal health of piglets challenged with E. coli F4 (1 mL, 106 CFU/mL). In total, 270 weaned piglets were used in a randomized block design with six treatments: positive control, with 40 mg/kg colistin (PC); negative control, without the growth promoter (NC); negative control +5 g/kg benzoic acid (BA); negative control +2 g/kg combination of BA+EO (BA+EO2); negative control +3 g/kg combination of BA+EO (BA+EO3); negative control +4 g/kg combination of BA+EO (BA+EO4). BA+EO3 presented a greater average daily gain (ADG) (p = 0.0013) and better feed-to-gain ratio (p = 0.0138), compared to NC, from 21 to 35 days age. For the total period, BA, BA+EO3, and BA+EO4 were similar to PC and superior to NC for ADG (p = 0.0002) and final body weight (BW) (p = 0.0002). No difference (p > 0.05) was verified for diarrhea, microbial population, production of volatile fatty acids, pH, weight of organs, cellular proliferation, and cholecystokinin count. NC and BA+EO4 resulted in a higher villus height in the jejunum (p = 0.0120) compared to BA+EO3. The use of BA or the combination of BA and EO at 3 g/kg provides improved performance, aside from being an economically viable alternative to replace colistin. Full article

This study investigated the potential of a Bacillus licheniformis-fermented feed additive (BLF) as an antibiotic substitute in weaning piglets. Ninety-six crossbred piglets were randomly allotted into four treatments with three replicate pens per treatment and eight pigs per pen. Piglets were fed diets as follows: a basal diet as control, a basal diet supplemented with bacitracin (30 mg/kg of bacitracin methylene disalicylate), a basal diet supplemented with BLF (1 g/kg of the Bacillus licheniformis-fermented feed additive), and a basal diet supplemented with bacitracin and BLF (15 mg/kg of bacitracin methylene disalicylate and 0.5 g/kg of the Bacillus licheniformis-fermented feed additive). The results showed that replacing all or half the bacitracin with BLF both reduced the incidence of diarrhea in weaning piglets from day 1 to 14. Principal coordinates analysis and a species abundance heat map showed that distinct clusters were formed between groups. Replacing all the bacitracin with BLF reduced bacterial evenness in the cecal digesta of weaning piglets, while the inhibitory effect on bacterial evenness was reversed in the group treated with bacitracin in combination with BLF. These results indicated that the half replacement of bacitracin with BLF was able to decrease the incidence of diarrhea and modify cecal microbiota composition in weaning piglets, suggesting that a Bacillus licheniformis-fermented feed additive has good potential as a suitable alternative to antibiotics use in the swine industry. The objective was to evaluate the effect of oral administration of Lactobacillus plantarum CAM6 strain as an alternative to antibiotics in weaned pigs on productive parameters, blood biochemical profile, and IgA serum levels. Thirty-six 21-day-old weaned piglets were randomly assigned to three groups with three replicates of four piglets each. Treatments consisted of a basal diet (BD; T0) without probiotics or antibiotics; BD + antibiotics and the same basal diet used in T0 plus oral administration of 5 mL × 109 CFU/mL of L. plantarum CAM-6 (T2). During the study (21 to 49 days of age) T2 obtained a similar live weight, weight gain, and feed conversion ratio when compared to the T1. Both treatments were better in these variables compared to T0 (p ≤ 0.05). Furthermore, T2 increased serum IgA levels (p ≤ 0.05). Additionally, hematological parameters and acid-base balance remained similar in all groups. However, significant reductions in the mean corpuscular hemoglobin concentration, platelets, and metabolic hydrogen ions were observed in T1 (p ≤ 0.05). The results of this study suggest that supplementation with L. plantarum CAM6 can be an alternative to antibiotics. Studies to evaluate its efficacy under commercial conditions and water administration require further evaluation. Full article

Three lactic acid strains were isolated from feces of the native Zungo Pelado breed of pigs (n = 5) and presumably identified as belonging to the Lactobacillaceae family by morphological techniques showing that they were Gram-positive/rod-shaped and catalase- and oxidase-negative. They were then identified by biochemical tests using API 50CHL as Lactobacillus plantarum (CAM6), Lactobacillus brevis (CAM7), and Lactobacillus acidophilus (CL4). However, 16S rRNA identification showed that all three strains were Lactobacillus plantarum. Additionally, all three isolates were able to grow in pH 3 and 4. Interestingly, the growth of the CAM7 strain decreased at pH 5.6 compared to that of the CAM6 strain (p < 0.05), and the growth of the CL4 strain was reduced at pH 7(p < 0.05). All three candidates showed good growth on bile salts (≥0.15%), and CAM6 and CAM7 showed better tolerance at higher concentrations (0.30%). Similarly, all strains tolerated sodium chloride (NaCl) concentrations from 2 to 10%. These strains also grew well at all temperatures tested (30, 37, and 42 °C). The CAM6 strain showed in vitro antibacterial activity against selected enteropathogenic bacteria (Escherichia coli strain NBRC 102203 and Salmonella enterica serovar Typhimurium 4.5.12) and commensal bacteria (Klebsiella pneumoniae ATCC BAA-1705D-5 and Pseudomonas aeruginosa ATCC 15442) and resistance to all antibiotics except amoxicillin. Further studies to evaluate the effects of these probiotic candidate strains in commercial pigs are currently underway. Full article

Zinc oxide (ZnO) has negative environmental effects and bioavailability in weaned piglets. Thus, finding safe and effective ZnO substitutes to improve intestinal health and to prevent diarrhea of weaned piglets is urgently required. Therefore, this experiment was conducted to evaluate the effects of hydrolysable tannins (HT), ZnO and HT versus ZnO on growth performance, antioxidant status, serum immunity, intestinal morphology, and digestive enzyme activities in weaned pigs. A total of 144 piglets (28 d-old, initial body weight 7.81 ± 0.99 kg) were assigned to 4 treatments with 6 replicates of 6 piglets each. The experiment lasted 28 d (d 1 to 14 as for phase 1 and d 15 to 28 as for phase 2). The dietary treatments include a corn-soybean meal basal diet (CON); ZnO diet (CON + 2000 mg/kg ZnO in phase 1 and 137.5 mg/kg ZnO in phase 2); HT diet (CON + 1000 mg/kg HT in the overall period (d 1 to 28); HT + ZnO diet (CON + 2000 mg/kg ZnO + 1000 mg/kg HT in phase 1, and 137.5 mg/kg ZnO + 1000 mg/kg HT in phase 2). In phase 1, the incidence of diarrhea was lower (p < 0.05) in the HT + ZnO group than CON. Serum catalase (CAT) and glutathione peroxidase (GSH-Px) were increased (p < 0.01) and malondialdehyde (MDA) was decreased (p < 0.01) in the HT + ZnO group than CON. Compared with CON, immunoglobulin M (IgM), immunoglobulin A (IgA) were increased (p < 0.05) in the HT + ZnO group. In phase 2, both HT and HT + ZnO had a trend to improve (p < 0.10) daily gain. The concentration of total antioxidant capacity (T-AOC) and IgM in serum was higher (p < 0.01) in HT compared with CON. Supplementation of HT improved (p < 0.01) GSH-Px activities in ileum mucosa than the ZnO group. Compared with CON, trypsin, lipase activities, and villus height of jejunum were improved (p < 0.05) in HT and HT + ZnO. The ratio of villus height to crypt depth in the jejunum was improved (p < 0.05) in the HT + ZnO group and which also was increased (p < 0.05) in ileum in the HT group compared with CON. Propionic acid, butyric acid, and acetic acid concentrations in the colon were increased (p < 0.05) in the HT group than CON. Overall, HT + ZnO treatments could be used to replace ZnO for reducing diarrhea and improving antioxidant capacity, immunity, and digestive enzyme activities in weaned piglets. Full article

Over the past decade, prebiotics have been used as an alternative approach in order to reduce the use of antimicrobials, by positively affecting the gut microbiota and decreasing the onset of several enteric diseases in pig. However, dietary supplementation with seaweed polysaccharides as prebiotics has gained attention in recent years. Seaweeds or marine macroalgae contain several polysaccharides: laminarin, fucoidan, and alginates are found in brown seaweeds, carrageenan in red seaweeds, and ulvan in green seaweeds. The present review focuses on studies evaluating dietary seaweed polysaccharide supplementation in pig used as prebiotics to positively modulate gut health and microbiota composition. Full article

During the prenatal, neonatal, and weaning periods, the porcine gastrointestinal tract undergoes several morpho-functional, changes together with substantial modification of the microbial ecosystem. Modifications of the overall structure of the small intestine also occur, as well as a rapid increase of the volume, mainly in the last period of gestation: intestinal villi, starting from jejunum, appears shortly before the sixth week of gestation, and towards the end of the third month, epithelial cells diversify into enterocytes, goblet cells, endocrine, and Paneth cells. Moreover, in the neonatal period, colostrum induces an increase in intestinal weight, absorptive area, and brush border enzyme activities: intestine doubles its weight and increases the length by 30% within three days of birth. During weaning, intestinal environment modifies drastically due to a replacement of highly digestible sow milk by solid feed: profound changes in histological parameters and enzymatic activity are associated with the weaning period, such as the atrophy of the villi and consequent restorative hypertrophy of the crypts. All these modifications are the result of a delicate and precise balance between the proliferation and the death of the cells that form the intestinal mucosa (i.e., mitosis and apoptosis) and the health conditions of the piglet. An in-depth knowledge of these phenomena and of how they can interfere with the correct intestinal function can represent a valid support to predict strategies to improve gut health in the long-term and to prevent weaning gut alterations; thus, reducing antimicrobial use. Zinc oxide (ZnO) at pharmacological doses is extensively employed in the pig industry as an effective tool to manage post-weaning diarrhea (PWD), a condition that causes huge economic losses because of its impact on the most pivotal phase of a piglet’s production cycle. In a multifactorial way, ZnO exerts a variety of positive effects along the entire gastrointestinal tract by targeting intestinal architecture, digestive secretions, antioxidant systems, and immune cells. ZnO also has a moderate antibacterial effect against Escherichia coli F4 (K88), the main causative agent of PWD. However, the environmental impact of ZnO and new emerging threats are posing serious questions to the sustainability of its extensive utilization. To work towards a future free from pharmacological ZnO, novel nutritional approaches are necessary, and many strategies have been investigated. This review article provides a comprehensive framework for ZnO utilization and its broad mode of action. Moreover, all the risks related to pharmacological ZnO levels are presented; we focus on European institutions’ decisions subsequently. The identification of a novel, complete solution against PWD should be accompanied by the adoption of holistic strategies, thereby combining good management practices to feeding approaches capable of mitigating Escherichia coli F4 (K88) infections and/or lowering ZnO utilization. Promising results can be obtained by adjusting diet composition or employing organic acids, natural identical compounds, polyphenol-rich extracts, prebiotics, and probiotics. Full article
