Title: Antifungal Activity of Cell-Free Filtrate of Probiotic Bacteria Lactobacillus rhamnosus ATCC-7469 Against Fungal Strains Isolated from a Historical Manuscript

This study was analyzed the cell-free filtrate (CFF) of the probiotic bacterial strain Lactobacillus rhamnosus ATCC-7469 28 to inhibit fungal growth isolated from a Historical Manuscript. Some comments should be in considered:

1-     1. Additional information about methods of enzymes and their activities should be included.

2-     2- What does this statement in line 217 of the MTT assay mean? After that, the growing cells were treated with a double-fold concentration of crude extract” Did the authors do this experiment using crude extract? What kind of crude extract is utilised?

3-     Add the untie of enzymes in figure 5

4-     What procedures were used to create probiotic bacteria metabolites? (Additional details to clarify the amount utilized in tissue culture).

Comments

It is a good paper and merits the publication as the authors  used multiple techniques to prove their study. However, the following minor comments need to be addressed before publication:

1. The number of references should be kept to a minimum and authors should omit older ones.

2. English language check is required as many grammatical errors should be corrected.

In their manuscript, the authors tested the antifungal activity of cell free extract of Lactobacillus rhamnosus against fungal strains that were isolated from the deteriorated historical books, while the scope of this manuscript is interesting but I have the following concerns

1- What the goal of this work, they have to cover that in the introduction

2- They did not explain that much about the nature of the active ingredients in the CFF (They seem to be fermentation products or secondary metabolites of acids and alcohols or esters)

3- The compounds identified in the CFF may have by themselves deterioration effects for the paper if we think from applied point of view, how do the authors configure the application of the CFF for this purpose

4- Figure 2, I do not see the arrows that indicate microbial growth

5- What  is the mechanism of action of the CFF

6- The authors have to explain statistical analysis in figure legends

7- Please check the language and grammar of the manuscript
