Special Issue Information

Dear Colleagues,

Malignant tumors have unique metabolic demands, which are required for growth under low oxygen but can also serve as vulnerabilities. These include metabolic adaptations to respiration with low oxygen. As respiration is required for tumor growth, even with oxygen limitations, tumors have adapted to alternative forms of respiration. Known adaptations include using alternatives to glucose to generate ATP, including glutamine and fatty acid oxidation, and providing intermediates that can be used in cellular respiration. Other adaptations include metabolic plasticity between respiration and glycolysis. Interventions that limit the plasticity of metabolism may render solid tumors unable to grow under hypoxic conditions, and certain respiratory adaptations, such as Sox2 expression, may govern the ability of tumors to metastasize or differentiate. Metabolic characteristics of cancer stem cells will be explored, including use of alternative substrates to glucose and respiratory adaptations that distinguish cancer stem cells from normal stem cells and normal respiration will be of particular interest.
