Round 1

Reviewer 1 Report

I read carefully the manuscript entitled: “Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production” and believe that it presents experiments of a good quality with relevant analyses. The research concerned screening analyzes aimed at the isolation of yeast strains with possible potential for the production of bioethanol from molasses. Appropriate research methods were used in the research to guarantee the identification of microorganisms. One significant drawback is the lack of tests on liquid media that would allow the evaluation of the efficiency of the process of converting sugars into ethanol by the isolated strains. The publication is recommended, subjected to minor revision.

However, here are few points which must be considered during revision of the manuscript:

# In the introduction, it is necessary to clearly indicate the novelty of the research in relation to the discussed research problem. The purpose of the research should be explained and the source of the microorganisms should be discussed in detail (justify the choice).

# The plate studies should be extended in the subsequent stages of the studies on liquid media cultures in larger volumes. An extremely important parameter that guarantees the possibility of using the isolated strains in industrial practice is the efficiency of ethanol production in conditions similar to practical ones.

# In my opinion, the results of research on the effectiveness of sugar conversion into ethanol by isolated yeast strains should be added.

To Reviewer 1-on-Duty

We would like to submit our manuscript revision entitled “Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production ”. Here are the details of the changing on the manuscript: we have edited the introduction section in the manuscript by clearly stating the purpose, novelty, and further reason of coconut water become the source of the microorganism. The design and method were explained in the abstract, introduction and method section. The research is focusing in the  isolation and identification to obtained novel yeast. The yeast isolation finalized by DNA sequencing method. While  the purpose of phenotypic test using solid media is only to see the yeast isolate could survive the stress conditions (high temperature, high acidity, and high molasses concentration).

We edited the manuscript using the mark “Track Changes” in Ms Word so the reviewer can see the changing.  We would be very happy if our revised manuscript could be acceptable in publication in internationally reputed journal “Fermentation”.

Reviewer 2 Report

A study of Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production: A few points deserve attention for further publication. I suggest that it is accepted for publication after the following revisions:

 - The authors could clarify in the abstract of the manuscript the mechanism, advantages, problems, and solutions for the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production.

 - In addition, authors should highlight the advantages/disadvantages of this Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production for industrial application and how this information will be addressed in the manuscript.

- Advantages for the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production: Which methods have advantages? Are they simple methods of contribution? When compared with other sustainable techniques? Authors need to leave this clear information throughout the text and the methods discussed in this manuscript. In addition, this information is needed for the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production systems contribution protocols are applied on an industrial scale.

- Problems with the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production systems: Does this protocol have a significant problem? This discussion could be improved.

- Additionally, advances in the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production systems with engineered tailor-made have been performed with other strategies. May open new opportunities. This discussion could be improved.

- This review had broached interest in the progress and recent applications of the Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production: The main contributions to the accomplishment of this work must be included in the conclusion.

 - Please, check all references according to the author's instructions.

- The manuscript must be formatted according to the journal's standards.

We would like to submit our manuscript revision entitled “Isolation of Novel Yeast from Coconut (Cocos nucifera L.) Water and Phenotypic Examination as the Potential Parameters in Bioethanol Production ”. Here are the details of the changing on the manuscript: we have clarified the problem, solution, and mechanism in the manuscript by changing and edited the statement in the abstract. The design and advantages of the method were explained in discussion section in paragraph 4 and 6.  The contribution of the research result stated in conclusion section. All the references already checked and relevant to the manuscript. The manuscript written by following the standard of fermentation journal.

We edited the manuscript using the mark “Track Changes” in MS Word so the reviewer can she the changing.  We would be very happy if our revised manuscript could be acceptable in publication in internationally reputed journal “Fermentation”.
