Reviewer 2 Report
This is a very well-written review paper studying the CatSper channels from different angles of sperm physiology and patophysiology. Such manuscript has an important merit for the understanding of sperm behavior and possible development of strategies for the management of male sub- or infertility. I have no major issues nevertheless, it would be interesting if the authors could add a paragraph on other ion channels CatSper may interact with during sperm activation and how are these interactions important on the downstream molecular machinery CatSper regulates. Also, I would recommend to specify what species (human, mouse, etc.) was the target model in the studies that are discussed in this review. This has been done for the most part, however the information is missing in several sections (for example lines 181-185, lines 310-313, lines 318-322, lines 350-353, etc.).

Response 1: We would like to extend our sincere thanks to your review for the insights and thoughtful suggestions. Taking those comments seriously, clarifications/corrections are made in the revised version (the modified content had been marked in red). We are glad that the quality of manuscript has been greatly improved and wish the revised manuscript meets the standards of approval.
Point 2: I have no major issues nevertheless, it would be interesting if the authors could add a paragraph on other ion channels CatSper may interact with during sperm activation and how are these interactions important on the downstream molecular machinery CatSper regulates.
Response 2: We really appreciate for your valuable comments. Following your suggestion, we have added a paragraph in the section of “overview of CatSper” to describe the cooperation of CatSper with other ion channels during sperm activation and how these ion channels regulate Catsper. Please see the details in the revised manuscript (line 95-112).
Point 3: Also, I would recommend to specify what species (human, mouse, etc.) was the target model in the studies that are discussed in this review. This has been done for the most part, however the information is missing in several sections (for example lines 181-185, lines 310-313, lines 318-322, lines 350-353, etc.).
Response 3: We would like to extend our sincere thanks to your valuable comments. According to your suggestion, we have specified what species (human, mouse, etc.) was the target model in the studies. Please see the details in the revised manuscirpt (line 178, 188, 191, 194, 200, 204, 218, 225, 319, 328, 359, 363, 389, 390, 402, and 427).
