Inhibitors for Alkaline Phosphatase

Phosphatase is an enzyme that uses water to cleave a phosphoric acid monoester into a phosphate ion and an alcohol. Phosphatase enzymes catalyze the hydrolysis of their substrates, making them subcategories of hydrolases. In cellular regulation and signaling, phosphatases and protein kinases play essential roles in phosphorylation and dephosphorylation (e.g., protein kinases). To better understand the role of alkaline phosphatase (ALP) in health and disease, it is imperative to develop small drug-like molecules that selectively inhibit ALP. Theophylline and (6S)-6-phenyl-2,3,5,6-tetrahydroimidazo thiazole, commonly known as levamisole, are well-known inhibitors of ALP. Similarly, the selenium analogue of tetramisole(selenotetramisole) was also synthesized and tested for its ALP inhibition activity. A commonly used anticonvulsant barbiturate drug, phenobarbital [5-ethyl-5-phenyl-2,4,6(1H,3H,5H)-pyrimidinetrione], was found to be an effective drug for the target of ALP.

This Special Issue on “Inhibitors for Alkaline Phosphatase” will cover synthesized compounds for the target of ALP and their role toward this protein.
