Round 1

Reviewer 1 Report

The manuscript by Tiz and co-workers submitted for publication in "Pharmaceutics" describes 15 small molecule drugs approved in 2022 by American Food and Drug Administration and, for each of them, provides the biological activity and the synthetic strategy. Overall, the current manuscript is well organized and will be of interest to others in this field and hence I support for pubblication in "Pharmaceutics".

Author Response

As the referee did not raise any point, we thank him/her for the appreciation

Reviewer 2 Report

The review by Tiz et al. of FDA approved drugs in 2022 is well written, informative, and highly readable. The review provides an introduction to a drug's mechanism of action followed by a discussion of its synthetic route. The authors also highlighted some trends in the 2022 FDA approval, such as approval of drugs bearing the sulfonamide scaffold, incorporation of deuterium, combination drugs, etc. I have one comment - please include the mechanism of action for ganaxolone.

Author Response

As referee suggests we included two lines highlighting a possible ganaxolone mechanism of action, in addition two further references were added, new 17 and 18.
