Round 1

Reviewer 1 Report

The present review on the use of excipients in the pediatric population and it's safety issue, is well written and easy to read. Moreover, the authors eaxustively give to the readers  the actual dimension of this problem and the need of further studies given the little information regarding excipients in pediatric medicines.

It’s originality is rapresented by the addressed topic itself that, usually, is not properly considered. In fact, most of the scientific literature is mainly focused on tolerability issues about drugs commonly used, both in- and off-label, in the pediatric population, I never red about concerns about the potential adverse effects directly associated to excipients rather than those of active substances in pediatrics. Obviously, some excipients are mandatory to obtain medicine suitable for children. However, using reccomended  amount or choosing alternative formulation could represent usefull strategy to reducing risks associated to these substances. Moreover, this manuscript highlighted also the further effort that the regulatory agencies must reserve to this issue.
